为了明确镉(Cd)修复微生物菌剂的构建原理及其作用效果,选取从超积累植物东南景天根内分离得到的5种植物促生菌,通过菌株共培养试验确定了作用效果最好的混合菌株组合,与富集植物油菜构建植物-微生物修复技术体系,在此基础上测定了油菜生物量、根系发育、Cd吸收积累、过氧化氢含量、谷胱甘肽含量以及抗氧化酶活性等。结果表明:芽孢杆菌Bacillus sp.Bac1和伯克霍尔德菌Burkholderia sp.Bac2的共培养生长状况最好,更适用于作为Cd修复菌剂。此外,接种不同浓度配比的Cd修复菌剂的作用效果有所不同。当Bac1和Bac2菌剂浓度配比为3∶1时更有利于油菜生长发育及对Cd的提取修复,而Bac1和Bac2菌剂浓度配比为1∶1时油菜植物抗氧化酶活性、谷胱甘肽含量等抗氧化系统指标对Cd胁迫的响应更为强烈。 |

To elucidate the constructing principles of efficient Cd-remediation agents and its functional effects on oilseed rape, five PGPB strains isolated from the hyperaccumulating plant Sedum alfredii Hance were selected in this study, and the optimal mixed strain combinations were thereby determined by strain co-culture test, and combine it with the enrichment plant oilseed rape, to establish a plantmicrobial remediation system. After that, the effects of Cd-remediation agents inoculation with different concentration ratios on plant biomass, root development parameters, Cd uptake and accumulation, peroxide(H2O2) contents, glutathione(GSH) contents and antioxidative enzymatic activities were investigated. The results showed that the combination of the two PGPB strains, namely Bacillus sp. Bac1 and Burkholderia sp. Bac2 had the best co-culture growth condition and was more suitable as a Cd remediation agent. In addition, the functional effects of Cd-remediation agents on oilseed rape growth, Cd uptake, and accumulation varied due to the different inoculating concentration ratios of Cd-remediation agents. When the concentration ratio of Bac1 and Bac2 was 3∶1, inoculation of Cd-remediation agents was more favorable to the growth and development of oilseed rape and the Cd phytoextraction, while the antioxidant system of oilseed rape including antioxidative enzymatic activities, H2O2 contents, and GSH contents responded more strongly to Cd stress when the concentration ratio of Bac1 and Bac2 was 1∶1. |
