珠江河网淡水鱼、虾和河蚬重金属污染特性及安全性评价

摘要: 为了全面评价珠江河网水产品重金属残留及食用安全风险,对鱼、虾和河蚬重金属的含量进行了检测分析,同时采用单因子污染指数(Pi)、综合污染指数(MPI)和健康风险评价模型对其污染程度、食用致癌和非致癌风险进行了评价。结果表明,水产品中重金属的含量(以鲜质量计)范围(均值)分别为Cr 0.031~0.264(0.131)mg·kg-1、Ni 0.077~0.742(0.170)mg·kg-1、Cu 0.199~22.575(2.318)mg·kg-1、Zn 3.422~36.764(7.939)mg·kg-1、As 0.112~4.192(0.339)mg·kg-1、Cd 0.004~1.269(0.095)mg·kg-1、Hg 0.012~0.048(0.026)mg·kg-1、Pb 0.028~0.253(0.096)mg·kg-1。鱼类和虾样品重金属残留均在安全值以内,但河蚬As和Cd残留略超标准值。水产品重金属污染程度评价结果表明,单因子污染指数排序为Cd > As > Pb > Hg > Cr > Zn > Cu。MPI值表明,不同水产品的污染程度依次为贝类(1.038)>虾类(0.353)>鱼类(0.101~0.292)。不同水产品重金属残留量的差异主要与它们不同的摄食习性、生活环境和对特定重金属的富集能力有关。健康风险评价结果表明,水产品复合重金属总目标危害系数(TTHQ)的高低顺序依次为河蚬>虾>鳢>鲶鱼>鲫鱼>翘嘴红鲌>鲈鱼>麦鲮>鲤鱼>餐条>罗非鱼>鲢鱼>广东鲂>鲮鱼>草鱼>赤眼鳟>鳙鱼。复合重金属TTHQ大于1,其中As的贡献比例最高,平均贡献率为51.7%。研究表明,当地居民若长期食用河蚬存在一定致癌风险。

Pollution status and safety assessment of heavy metals in natural fish, shrimp, and shellfish from Pearl River waterway

Abstract: In order to comprehensively evaluate residual, ecological, and edible health risks of natural aquatic products caused by heavy metals in the Pearl River waterway, the concentrations of heavy metals in fish, shrimp, and shellfish were determined. The extent of pollution carcinogenic and non-carcinogenic risks of aquatic products were evaluated by single factor pollution index (Pi), comprehensive pollution index (MPI), and various health risk assessment models. The results showed the following concentration ranges (means) of heavy metals in aquatic products in fresh weight:Cr 0.031~0.264 (0.131) mg·kg-1, Ni 0.077~0.742 (0.170) mg·kg-1, Cu 0.199~22.575 (2.318) mg·kg-1, Zn 3.422~36.764 (7.939) mg·kg-1, As 0.112~4.192 (0.339) mg·kg-1, Cd 0.004~1.269 (0.095) mg·kg-1, Hg 0.012~0.048 (0.026) mg·kg-1, Pb 0.028~0.253 (0.096) mg·kg-1. Compared with different aquatic products, the metal concentrations in fish and shrimp samples were within the quality safety limits, but the As and Cd concentrations of shellfish slightly exceeded the corresponding limits. The single factor pollution index of the extent of heavy metal pollution of aquatic products was in the order Cd > As > Pb > Hg > Cr > Zn > Cu. The MPI values showed that the degree of pollution of different aquatic products was shellfish (1.038) > shrimp (0.353) > fish (0.101~0.292). The heavy metal residues in different fishes were mainly attributed to their different feeding habits, living environment, and accumulation ability to specific heavy metals. In health risk assessment, the order of TTHQ (Total target hazard quotient) of aquatic products was as follows:shellfish > shrimps > Eleotridae > catfish > crucians > Culter alburnus > perch > mrigal carp > carp > sharpbelly > tilapia > silver carp > Gungdong bream > dace > grass carp > Barbel chub > bighead fish. The TTHQ of complex heavy metals was more than 1, and the contribution rate of As was the highest with an average contribution rate of 51.7%. Thus, there is a certain risk of carcinogenesis associated with edible shellfish.

珠江三角洲位于广东省的东南部,地处南方亚热带区域,毗邻中国南海,雨量充沛,水资源丰富,河网水系纵横交错,是我国南方城市和人口最集中的地区之一。随着经济快速发展,工业化和城市化加速,重金属等有毒有害物质随生活、工农业废水直接或间接排放进入河流及水域,由于其在水环境中残留具有持久性,且不能被微生物降解[1],在某些条件下(如水环境pH和氧化还原电位变化),能通过食物链在不同食物层级水生生物中累积[2-3]。重金属污染一方面对水生生物造成直接毒害,同时也能通过人们对水产品的摄取,进入人体内造成现实和潜在健康风险[4-5]。据研究,长期摄入含重金属污染水产品,能导致许多不良影响,如肾功能受损、生殖能力下降、肝损伤、皮肤癌和膀胱癌,甚至死亡[6]。大量相关研究显示珠江三角洲地区水产品中存在不同程度的重金属残留,如梁辉等[7]于2010—2014年采集广东市售1326份水产品检测发现Pb超标率为0.15%,Cd超标率为5.35%;广东沿海近江牡蛎、珠江口鱼虾也都存在重金属超标现象[8]。水产品中含有丰富的蛋白质、不饱和脂肪酸、维生素和微量元素,对膳食营养均衡起到了关键的作用[9-10]。随着人们生活水平的提高,水产品在我国食品消费中所占比例日益提高,我国已成为水产品消费大国[11]。然而,随水环境污染的加重,水产品也成了环境污染物吸附和富集的载体,水产品质量安全和养殖环境污染风险评估受到社会广泛的关注[12]。但目前,珠江河网淡水鱼类、虾和河蚬重金属污染特征及其健康风险的研究尚鲜有报道,为此本研究以珠江河网淡水鱼类、虾和河蚬为研究对象,分析鱼类、虾和河蚬体内重金属的含量水平,并探讨重金属在不同水产品中的污染特征,评估当地居民食用水产品可能造成的健康风险,以期为珠江河网重金属污染防治、生态环境保护提供基础数据及科学依据。

1 材料与方法 1.1 研究区域和样品采集

珠江是我国第二大河,主要由西江、东江和北江三条支流组成。北江和东江在珠江三角洲交汇,形成了一条密集的水道,经8个出海口汇入南海。2016年1—5月,于21个地点进行了样品采集(见图 1)。样品包括淡水鱼、虾和河蚬,采样点分为四个采集区域,分别为上游采样区域(U1~U6)、中部采样区域(N1~N7)、东部河口区(S1~S4)和西部河口区(S5~S8),不同区域样品分别采集,鱼样取样量为4~6条鱼背部肌肉,虾为15尾、河蚬为30个,去壳后取可食部分,样品混匀后均质保存。具体各采样区域采集样品种类、性状和数量见表 1

1.2 仪器与试剂

仪器:Agilent 7500电感耦合等离子体质谱仪(ICP-MS,美国安捷伦公司)。

试剂:浓硝酸(德国Merck公司),30%双氧水优级纯(德国Merck公司),多金属元素混合标准溶液(GNM-0083,国家标准物质网),鱼肌肉标准参考物质(ERM-BB422,国家标准物质网)。

1.3 样品的预处理

称取2 g(鲜质量)样品于四氟乙烯压力消解罐中,加入10 mL浓硝酸(65%)浸泡过夜,再加入2 mL H2O2(30%),盖紧不锈钢外罐,置于恒温干燥箱中,将温度设定在120 ℃后,保持4 h,消解完毕,在箱内自然冷却至室温。取出内罐沸水浴加热1 h左右,赶酸,后将消解液移至25 mL容量瓶中,用去离子水定容至刻度,摇匀待测。

1.4 分析与质量控制

样品分析根据USEPA(美国环保局)3051A和200.8方法进行测定[13-14]。质量保证和质量控制(QA/QC):每批样品进行试剂空白、样品重复和样品加标检测,结果显示,所有样品的相对标准偏差(RSD)均小于10%,样品回收率为80%~120%,Cr、Ni、Cu、Zn、As、Cd、Hg和Pb样品检出限分别为0.008 2、0.003 4、0.021、0.048、0.005 1、0.001 3、0.001 5 mg·kg-1和0.003 3 mg·kg-1

1.5 重金属污染评价

水产品中重金属的单因子污染指数(Pi)用于评价水产品中单项重金属污染[15-16],计算公式如下:

式中:Ci为水产品重金属测定值的均值,mg·kg-1Si为水产品重金属限量标准,mg·kg-1,参考《GB 2762—2017食品安全国家标准食品中污染物限量》 [17]和文献[18],Si取值为Hg 0.3 mg·kg-1、Cr 2 mg·kg-1、Cu 50 mg·kg-1、Zn 150 mg·kg-1、Cd 0.1 mg·kg-1、As 0.5 mg·kg-1、Pb 0.5 mg·kg-1;评价标准:Pi < 0.2为正常背景值水平;0.2≤Pi < 0.6为轻污染水平;0.6≤Pi < 1.0为中污染水平;Pi≥1.0为重污染水平[11, 19-20]

采用重金属综合污染指数(MPI)比较了水产品重金属污染总量的差异[21-22]。计算公式如下:

式中:Cn为第n种水产品中测得重金属的含量平均值。

1.6 人体健康风险评估

分别采用USEPA使用的评价模型,对水产品非致癌[23-24]和致癌风险[25-26]进行评价。水产品非致癌风险以目标危害系数(THQ)为基础,对水产品摄入引起的非致癌有害影响的健康风险进行评估,其中较高的THQ反映出较高的长期非致癌影响概率,低于1的THQ值不反映任何明显的风险,当THQ值大于1时,暴露人群存在健康风险。计算公式为:

式中:C为水产品中重金属的浓度,μg·g-1EF为暴露频率(365 d·a-1);ED为暴露时间(70 a);FIR为食物摄取率,广东省居民的水产品摄取率为70.8 g·d-1(鲜质量)[27]WAB为人体平均体质量(60 kg);TA为平均暴露时间(ED×365 d);根据USEPA 2000年的数据,Zn、Co、Cu、Ni、Mn、Pb、Cd、Cr、As、Hg的RfD分别为0.3、0.04、0.000 3、0.12、0.02、0.004、0.001、1.5、0.000 3 mg·kg-1和0.000 1 mg·kg-1

复合重金属总目标危害系数TTHQ=∑THQ。

致癌风险评价公式为:

式中:Rigc是通过食物摄入重金属致癌的年平均风险值,a-1Dig为重金属经食入途径单位体质量日均最大暴露剂量,mg·kg-1·d-1qig为重金属摄入量致癌强度系数。Cd、As和Cr致癌强度系数的qig值分别为6.1、15、0.91[28],平均预期寿命为70 a。

采用公式(5)计算重金属日平均最大暴露剂量(Dig)。

式中:Dig为每单位体质量重金属通过摄食途径的最大日暴露剂量,mg·kg-1·d-1Iad为广东省居民水产品日平均摄入量(70.8 g·人-1·d-1,鲜质量);60为人均体质量,kg。

1.7 数据统计

使用Origin 6.0和Excel 2007进行数据统计分析。

2 结果与讨论 2.1 珠江三角洲河网水产品中重金属分布特征

珠江三角洲河网17种水产品(鱼类、虾和河蚬)样品检测结果(表 2)显示,重金属的含量范围和均值,分别为Cr(0.031~0.264,0.131)、Ni(0.077~0.742,0.170)、Cu(0.199~22.575,2.318)、Zn(3.422~36.764,7.939)、As(0.112~4.192,0.339)、Cd(0.004~1.269,0.095)、Hg(0.012~0.048,0.026)、Pb(0.028~0.253,0.096)mg·kg-1(鲜质量)。Zn与Cu为鱼类必需元素,参与鱼体的新陈代谢过程,更易被鱼体主动吸收,在检测样品中残留量较高[29],与鱼体本身残留和较强的吸收能力有关。Cr、Ni、As、Cd,Hg和Pb为非必需元素,不参与生理活动,进入体内能产生较强的毒性[30],国内外对其有严格的安全限量标准[31]。对鱼类、虾和河蚬重金属残留进行比较,根据《GB 2762—2017食品安全国家标准食品中污染物限量》,鱼类和虾样品中所测金属残留均在安全值以内,但河蚬样品中As和Cd略超限量值,为安全值的1.3倍和1.2倍。鱼类中重金属含量虽未超过水产品中有毒有害物质的限量,但As和Pb在所有鱼类样品中仍然是高残留,平均残留量分别为安全标准值的65.4%和20.2%。

目前,我国不同区域水产品中都存在不同程度的重金属超标,如北京市市售4种鱼类中存在Pb、Cd和As 3种重金属超标,超标率分别为14.6%、5.2%和10.4%[11];台海浅滩渔场水产品Cd超标率为9.7%[32],广西刁江野生鱼类As、Pb超标率分别为64.56%、5.06%[3],钱塘江、赤水河、滦河等流域中的野生鱼类监测数据表明,Pb、Cr和Cd的污染程度较突出[33-35],在本研究中珠江三角洲河网主要鱼类重金属超标率并不明显。

2.2 鱼类、虾及河蚬肌肉中重金属污染程度及比较

采用单因素污染指数法对样品中7种元素的污染程度进行评价(图 2),64.4%~100%的样品Pi值均在正常背景值和轻度污染范围内。这表明珠江三角洲鱼类重金属污染相对较低。在被测样品中,Cr污染水平最低,只有一个样品残留浓度高于背景值。Cu、Zn、Hg的轻污染样品比例分别为6.78%、3.39%和4.24%,有轻微的微量污染。Pb的污染程度略高,轻度和中度污染样品的比例分别为22.90%和3.39%。Cd污染程度较高,中、重度污染样品比例为4.24%、5.90%。As污染最为严重,中度污染占23.73%,重度污染占11.86%。水产品重金属污染程度的单因子污染指数排序为Cd>As>Pb>Hg>Cr>Zn>Cu。利用复合重金属污染指数(MPI)比较了水产品重金属污染的总体差异,如图 2所示。MPI值表明,不同水产品的污染程度依次为河蚬(1.038)>虾(0.353)>鳢(0.292)>鲶鱼(0.195)>鲈鱼(0.177)>鲫鱼(0.170)>翘嘴红鲌(0.164)>鲤鱼(0.138)>鲮鱼(0.130)>鲢鱼(0.128)>麦鲮(0.126)≈餐条(0.126)>草鱼(0.121)>鳙鱼(0.120)>赤眼鳟(0.116)>广东鲂(0.109)>罗非鱼(0.101)。分析珠江河网河蚬、虾和鱼类重金属残留量,MPI存在一定的差异,河蚬MPI明显高于虾和鱼类,据相关的研究证实水环境中重金属在沉积物中含量最高,依次为底栖生物、鱼类,最低为水体,河蚬为底栖滤食性动物,相对于鱼类,其对重金属有更高的富集能力[36-37]。鱼类重金属残留量的差异同时也与它们不同的摄食习性有关,食肉鱼类由于处于食物链的较高层级,比食草鱼类和杂食性鱼对重金属富集能力更强。鳢、鲶鱼、鲈鱼主要是食肉动物,处于水生食物链的顶端,且生活在水体的中下层,具有较强的环境耐受性,因此重金属含量较高,而罗非鱼为杂食性鱼类,草鱼为植食性鱼类,重金属残留量相对较低。

2.3 鱼、虾和河蚬重金属健康风险评价

当地居民通过摄入水产品暴露重金属的THQ评价值如表 3所示。单一重金属的THQ从高到低的顺序依次为As>Cd>Ni>Cu>Hg>Mn>Pb>Zn>Cr。除As外,食用水产品其他单个元素THQ大都低于1。贝类、虾和鱼类复合重金属TTHQ的高低顺序依次为河蚬>虾>鳢>鲶鱼>鲫鱼>翘嘴红鲌>鲈鱼>麦鲮>鲤鱼>餐条>罗非鱼>鲢鱼>广东鲂>鲮鱼>草鱼>赤眼鳟>鳙鱼。复合重金属TTHQ均大于1,其中As的贡献比例最高,平均贡献率为51.7%,其次为Cu,平均贡献率为24.06%,Hg贡献率为11.92%,说明As是主要风险元素。

采用USEPA健康风险评价方法对珠江三角洲主要水产品中致癌元素As、Cr、Cd进行分析,结果见图 3。由图 3可知,As(1.76×10-4 a-1)致癌风险最大,Cd(3.05×10-6 a-1)次之,Cr(2.92×10-6 a-1)最低;国际辐射防护委员会(ICRP)推荐的通过饮食途径最大可接受年风险水平为5×10-5 a-1,所测水产品中As已略超致癌风险值(5×10-5 a-1),其中,河蚬通过食物摄入As致癌的年平均风险值为3.41×10-4 a-1,说明食入河蚬相较其他水产品有较高致癌风险。

3 结论

(1)根据《GB 2762—2017食品安全国家标准食品中污染物限量》,鱼类、虾样品中所测重金属残留均在安全值以内,但河蚬样品中As和Cd略超限量值,河蚬重金属浓度明显高于虾和鱼。重金属综合污染指数(MPI)结果显示,不同水产品的污染程度依次为河蚬(1.038)>虾类(0.353)>鳢(0.292)>鲶鱼(0.195)>鲈鱼(0.177)>鲫鱼(0.170)>翘嘴红鲌(0.164)>鲤鱼(0.138)>鲮鱼(0.130)>鲢鱼(0.128)>麦鲮(0.126)≈餐条(0.126)>草鱼(0.121)>鳙鱼(0.120)>赤眼鳟(0.116)>广东鲂(0.109)>罗非鱼(0.101)。不同水产品重金属残留量的差异主要与它们摄食习性、生活环境和富集能力有关。

(2)目标危害系数(THQ)评价结果显示,重金属在水产品中产生的健康风险大小依次为As>Cd>Ni>Cu>Hg>Mn>Pb>Zn>Cr。复合重金属总目标危害系数(TTHQ)结果表明,水产品健康危害风险大小依次为河蚬>虾>鳢>鲶鱼>鲫鱼>翘嘴红鲌>鲈鱼>麦鲮>鲤鱼>餐条>罗非鱼>鲢鱼>广东鲂>鲮鱼>草鱼>赤眼鳟>鳙鱼。当地居民若长期摄入河蚬和虾,暴露重金属的健康风险较大,As是主要风险元素。
