The toxicity effect of Cu2+-ofloxacin complex on Vero cell

Abstract:
      It is very important to study the growth inhibition of Cu2+-ofloxacin chelate on Vero by cytotoxicity test for understanding the toxic effects of the two kinds of pollutants on cells alone and in combination. The experiments were conducted in order to understand the toxicity effect of Cu2+-ofloxacin chelate on Vero. The inhibition rate of cell growth was calculated by MTT assay based on cultured Vero under normal culture conditions for 3 days and then cultured with OFL(0.63 mg·L-1, 1.25 mg·L-1, 2.5 mg·L-1, 5 mg·L-1 and 10 mg·L-1) and Cu2+(2.75 mg·L-1, 6.88 mg·L-1) treatment levels for 24 h. The results showed OFL and Cu2+ led to changes in the cell morphology, the cell disruption and decreases in adherence rate. The inhibition rate of Vero cell was increased with increases in the concentration of Cu2+. The effect of OFL on Vero cell growth was significant. The inhibition rate of Vero cell was between 30%~36% with the Cu2+-OFL compound at 2.75 mg·L-1 Cu2+. At the concentration of OFL was 2.5 mg·L-1, 5 mg·L-1 and 10 mg·L-1, significant difference was observed between the inhibition rate of Vero cell with the Cu2+-OFL compound and the sum of OFL or Cu2+ alone. But, significant changes in the inhibition rate of Vero cell was not observed with changes in OFL concentrations. At 6.88 mg·L-1 Cu2+, and 1.25 and 2.5 mg·L-1 OFL,significant difference was not observed between the inhibition rate of Vero cell with the Cu2+-OFL compound and the total of OFL or Cu2+ alone. When the concentration of OFL were 0.63, 5 and 10 mg·L-1, the inhibition rate of Vero cell of the Cu2+-OFL compound was lower than the sum of OFL and Cu2+ alone. The inhibition rate of Vero cell of the Cu2+-OFL compound increased with increases in OFL concentrations. The inhibition rate of Vero cell of the Cu2+-OFL compound was lower than the sum of the OFL and Cu2+ alone. OFL and Cu2+ can antagonize to the growth of Vero cell.
