Determination of 12 phenolic acids in straw extract or straw decomposed liquid through high performance liquid chromatography

Abstract:
      A method of reversed phase high performance liquid chromatography was established, which could be used to determine the contents of 12 phenolic acids(gallic acid, protocatechuic acid, gentisic acid, p-hydroxybenzoic acid, chlorogenic acid, vanilla acid, coffee acid, syringic acid, p-coumaric acid, ferulic acid, sinapic acid, salicylic acid) simultaneously in straw extract or decomposed liquid. Waters HLB(500 mg, 60 μm, 6 mL) solid-phase extraction cartridges were used to concentrate phenolic acids in aqueous solution. The gradient elution was carried out using acetonitrile solution containing 0.5% acetic acid and 0.5% acetic acid solution as the mobile phase. The diode array detector was used for detection, and the determination of 12 kinds of phenolic acids could be completed simultaneously in 30 minutes. All of phenolic acids could reach the baseline separation. The recovery rate of 12 phenolic acids were between 79.41%~101.92%, and the RSD of retention time and peak area of 12 phenolic acids were between 0.02%~0.1% and 0.25%~2.26%, respectively. This method is rapid, sensitive and accurate, suitable for the simultaneous determination of 12 phenolic acids existed in straw extract, straw decomposed liquid, soil solution and aqueous solution.
