Abstract:
      The hepatoprotective effects of Glycyrrhiza glabra extract(GGE) against TCDD-induced liver injury was evaluated using precision cut liver slices(PCLS). The PCLS were randomly divided into six groups: control group, TCDD model group, GGE group(0.8 mg·mL-1), GGE pre-treatment group, GGE post-treatment group, and GGE pre-and post-treatment group. To PCLS culture system 0.2, 0.4 and 0.8 mg·mL-1 GGE were added before(pre-treatment), after(post-treatment) and both before and after(pre-and post-treatment) exposure to 0.3 μg·L-1 TCDD. The culture supernatants of PCLS were collected for detecting activities of glutamic pyruvic transaminase(GPT), glutamic oxaloacetic transaminase(GOT), and superoxide dismutase(SOD), and contents of malondialdehyde(MDA), tumor necrosis factor-α(TNF-α), interleukin-1β(IL-1β) and immunoglobulin M(IgM). Results showed that the pre-treatment group had relieving effect on liver injury by TCDD. The GGE significantly inhibited the elevation of GOT, GPT, MDA,TNF-α,IL-1β, and IgM, but significantly increased level of superoxide dismutase(SOD). Overall results demonstrate the hepatoprotective effects of GGE and support the use of GGE as a hepatoprotective agent in fish.
